蒺藜苜蓿PHD-Finger家族鉴定和表达分析

摘 要:

PHD-Finger家族基因广泛存在于植物当中,在抵抗非生物胁迫过程中起到重要的作用。本研究鉴定到分布在蒺藜苜蓿8条染色体上的64个PHD-Finger家族基因。进化树分析显示蒺藜苜蓿的PHD-Finger家族基因分为14个亚家族,共有6个基因复制事件发生。基因结构分析发现蒺藜苜蓿PHD-Finger家族成员多数含有大量的内含子。通过RT-qPCR方法验证了MtPHD3MtPHD13MtPHD19MtPHD61在盐胁迫和干旱胁迫条件下转录水平上升,MtPHD12MtPHD18MtPHD19MtPHD38在冷胁迫条件下转录水平上升,暗示这些基因在苜蓿的胁迫应答中起重要作用。本研究将为深入研究蒺藜苜蓿PHD-Finger家族成员的基因功能和分子育种工作提供理论基础。

Genome-wide identification and expressional analysis of the PHD-Finger family in Medicago truncatula

Abstract:

PHD-Finger family are widely conserved in plants and play an important role in response to abiotic stress. In our study, 64 PHD-Finger family genes were identified on eight chromosomes in Medicago truncatula. Phylogenetic analysis indicated that the PHD-Finger family members could be grouped into 14 subfamilies. A total of 6 gene replication events occurred. Gene structure analysis indicate that most of the PHD-Finger family members in M. truncatula containve a large number of introns. The results of real-time quantitative PCR confirmed that the transcription levels of MtPHD3, MtPHD13, MtPHD19 and MtPHD61 were increased under salt and drought stress. And the transcription levels of MtPHD3, MtPHD12, MtPHD18 and MtPHD19 were increased under low temperature stress, suggesting that these genes play an important role in stress response in M. truncatula. This study will provide a theoretical basis for further study of gene function of PHD finger family members and molecular breeding M. truncatula in.
